Argireline (Acetyl Hexapeptide-8), Snap-8 (Acetyl Octapeptide-3), Syn-Ake (Dipeptide Diaminobutyroyl Benzylamide Diacetate) and Leuphasyl (Pentapeptide-18) are cosmetic amino acid sequences made to soften the look of expression lines, the creases that form where your face moves most. Each maker describes, from laboratory research, how its peptide was designed. Supplier use levels run from 1% to 10%, and the human studies behind them are small, so the concentration you use and the expectations you set both matter.
- Argireline: INCI Acetyl Hexapeptide-8, made by Lubrizol (Lipotec). Lubrizol lists 2 to 10% for Argireline peptide solution C and 2 to 5% for Argireline Amplified.
- Snap-8: INCI Acetyl Octapeptide-3, made by Lubrizol (Lipotec). Lubrizol lists a 3 to 10% use level.
- Syn-Ake: INCI Dipeptide Diaminobutyroyl Benzylamide Diacetate, made by dsm-firmenich. Distributor Univar lists 1 to 4% and pH 4.5 to 5.5.
- Leuphasyl: INCI Pentapeptide-18, made by Lubrizol (SpecialChem listing). Distributor Lotioncrafter lists 3 to 10%.
- Solubility and heat: the standard grades of all four are water-soluble and are added at cool-down, below 40°C (104°F), per Lotioncrafter and Lotioncrafter's Syn-Ake sheet.
What are expression lines?
Expression lines are the creases that form where facial muscles move over and over: forehead lines, the "11s" between the brows, crow's feet and the fine lines around the lips. Lubrizol describes them as "persistent expression wrinkles caused by facial muscle contractions" on its Snap-8 product page.
Lines driven mainly by dryness or sun are a different job. Collagen-signal peptides such as Matrixyl 3000 aim at the look of firmness. Expression line peptides aim at the look of lines that come from movement.
What are neurotransmitter-modulating peptides?
They are short amino acid sequences that their makers describe, from laboratory models, as working on the signaling steps behind facial movement. Each one is described as targeting a different step:
- Argireline and Snap-8 target the SNARE complex, the protein machinery that releases the signaling chemical. Lubrizol says Argireline "destabilizes the SNARE complex" on its Argireline page, and describes Snap-8 as a copy of the start of the SNAP-25 protein that competes for a place in that complex (SpecialChem).
- Syn-Ake works at the receiving end. dsm-firmenich describes it, from laboratory work, as acting at the muscle's acetylcholine receptor, where the signal lands. The maker says its sequence is modeled on Waglerin-1, a peptide found in temple viper venom; Syn-Ake itself is a lab-made amino acid derivative, not venom (dsm-firmenich).
- Leuphasyl is described as imitating the natural pathway of enkephalins, and a review describes it as damping nerve activity in laboratory research (SpecialChem; Schagen, 2017).
Read those as maker descriptions of laboratory research. On skin, the realistic goal is a change in how lines look.
What is Argireline (Acetyl Hexapeptide-8)?
Argireline is a six-amino-acid sequence, and Lubrizol calls it "the first acetyl hexapeptide-8 in the cosmetic market" (Lubrizol). Its current INCI name is Acetyl Hexapeptide-8. You may see it formerly listed as Acetyl Hexapeptide-3 on older labels and in older papers (CosIng record).
Lubrizol sells three grades, and they are not interchangeable:
- Argireline peptide solution C: Water (Aqua), Acetyl Hexapeptide-8, Caprylyl Glycol. Use level 2 to 10% (Lubrizol).
- Argireline Amplified peptide solution: Water (Aqua), Acetyl Hexapeptide-8, Sodium Benzoate. Use level 2 to 5% (Lubrizol). Lotioncrafter lists pH 4.0 to 5.5 as supplied and a formula working range of pH 4.0 to 8.0 (Lotioncrafter).
- Argireline YOUth peptide: Lubrizol describes it as an oil-soluble version (Lubrizol). It is the grade designed for oil-based products.
What are the benefits of Argireline, and what do the studies show?
The benefit people look for is a softer look to forehead lines and crow's feet. Here is what the published record holds:
- Lubrizol's own claim: a 20.6% reduction in the appearance of wrinkles in one week with a 2% solution. The product page does not list panel size or design (Lubrizol).
- Blanes-Mira and colleagues, 2002: the original paper on the peptide. A review summarizing it reports 10 women, a 10% cream twice daily for 30 days, and about a 30% improvement in eye-area lines (Schagen, 2017).
- Wang and colleagues, 2013: 60 adults in China, Argireline or placebo in a 3:1 split, twice daily for 4 weeks. Subjective scoring gave 48.9% efficacy against 0% for placebo, and silicone replicas showed lower roughness in the Argireline group (study record). A dermatologist's review points out that the study was not blinded (Baumann, MDedge).
- Henseler, 2023: 19 women, split-face and double-blind, twice daily for 4 weeks. Camera-measured wrinkle scores fell slightly on both sides, with no significant difference between the Argireline side and the control side (Henseler, 2023).
My read after 20 years of formulating: Argireline has the longest track record here, and the results are mixed. Expect a modest softening, not a transformation.
Does Argireline get into the skin?
Mostly it stays on the surface. In a lab study on guinea pig and human donor skin, a 10% emulsion left less than 1% of the peptide in the skin, most of it in the outer stratum corneum, and none was found in the dermis (Kraeling, 2015, as summarized by Baumann). A porcine skin study found that a water-in-oil-in-water emulsion moved more peptide into the outer layer than simpler emulsions. A dermatologist's review suggests the smoothing seen in some studies may come from better surface hydration, since another study measured lower water loss with Argireline (Baumann, MDedge).
What is Snap-8 peptide (Acetyl Octapeptide-3)?
Snap-8 is an eight-amino-acid sequence from the same maker, built as a longer relative of Argireline. Its INCI on the standard grade is Water (Aqua), Acetyl Octapeptide-3, Caprylyl Glycol, and Lubrizol lists a 3 to 10% use level (Lubrizol). It is water-soluble and goes in at cool-down, below 40°C (Lotioncrafter).
What are the benefits of Snap-8 peptide?
The target is the same: the look of expression lines around the eyes and forehead. The distributor sheet reports that the severity of lines around the eyes decreased by up to 63.13% in clinical studies, and that Snap-8 is about 30% more active than Argireline in lab and human work (Lotioncrafter). Neither the panel size, the length of the study nor the concentration appears on that sheet. These are supplier figures, and I could not find an independent study on Snap-8 to set beside them.
What is Syn-Ake?
Syn-Ake is the dsm-firmenich expression line peptide, sold as a glycerin and water solution: Glycerin, Aqua, Dipeptide Diaminobutyroyl Benzylamide Diacetate (dsm-firmenich). The INCI calls it a dipeptide derivative, while suppliers describe it as a tripeptide. When you shop, match the INCI name, not the nickname.
A Syn-Ake technical document issued by Pentapharm (Basel, Switzerland) gives a use level of 1 to 4%, a specification pH of 4.5 to 5.5, stability from pH 3.0 to 5.5, and compatibility with up to 50% ethanol (Pentapharm technical file). Supplier studies used 4%:
- 45 volunteers, 28 days, twice daily, Syn-Ake 4% against a placebo and against 10% Acetyl Hexapeptide-3. The supplier reports Syn-Ake gave the larger change in forehead smoothness and line depth (Pentapharm).
- The "up to 52%" figure that circulates online comes from one volunteer's forehead measurement in that same document. One person is not a result you can plan around.
- dsm-firmenich also describes a 100-volunteer study, 25 per group, 4% against placebo, twice daily for 4 weeks. The page does not list the numeric results (dsm-firmenich).
What is Leuphasyl (Pentapeptide-18)?
Leuphasyl is a Lubrizol (Lipotec) five-amino-acid sequence. Its INCI is Aqua, Glycerin, Pentapeptide-18, Caprylyl Glycol (SpecialChem). Lotioncrafter lists a use level of 3 to 10%, a pH of 2.0 to 5.0 as supplied, water solubility and cool-down addition below 40°C (Lotioncrafter).
Lotioncrafter describes Leuphasyl as complementing Argireline (Lotioncrafter). A review reports a 43-woman study at 0.05% each: Leuphasyl alone gave an 11.64% reduction in line depth, Acetyl Hexapeptide-3 gave 16.26%, and the two together gave 24.62% (Schagen, 2017). A separate 2014 study of 20 volunteers over two months found visible change only at the 2% level, with average line size down 34.7% on the forehead and 28.4% around the eyes. It had no placebo group (Dragomirescu, 2014).

How do you read the percentage on a peptide label?
This is the part most brands skip. These ingredients are sold as dilute solutions, and the percentage on a label almost always refers to the solution, not the pure peptide.
Lotioncrafter states its Snap-8 solution contains 0.05% Acetyl Octapeptide-3 (Lotioncrafter). So a product with "10% Snap-8" holds about 0.005% of the peptide itself. That is normal, and the supplier use levels are written for the solution. The point is to compare like with like: 10% Snap-8 solution against 10% Snap-8 solution, not against a figure for pure peptide.
Two more label checks:
- Look for the INCI name in the ingredient list. "Argireline" on the front with no Acetyl Hexapeptide-8 on the back is a question to ask the brand.
- Look for a stated percentage. At Skin Perfection we print exact concentrations and never hide actives in a proprietary blend, because this is the only way you can compare formulas.
How to use expression line peptides in a routine
Skin Perfection sells these as serum boosters, raw peptide solutions you add to a water-based serum, lotion or cream. They are not meant to go on skin undiluted. - Pick a water-based base. All four standard grades are water-soluble. For an oil-based product, Lubrizol offers the oil-soluble Argireline YOUth grade.
- Stay inside the supplier range. 2 to 10% for Argireline solution C, 2 to 5% for Argireline Amplified peptide, 3 to 10% for Snap-8 and Leuphasyl, and 1 to 4% for Syn-Ake. For example, 5% Snap-8 in 1 oz of serum is 45 drops.
- Mind pH. The Syn-Ake technical file lists stability from pH 3.0 to 5.5, so keep a Syn-Ake base inside that window.
- Keep it cool. Refrigerate the booster on arrival and blend it into a base at room temperature, never into a hot phase above 40°C.
- Apply where you move. Smooth the finished blend over the forehead, between the brows, around the eyes and around the lips, morning and night.
- Give it a month. The human studies above ran 4 to 8 weeks. Judge results with same-light photos at week 0 and week 4.

- Start with Argireline if you want the ingredient with the most published human data, positive and negative.
- Choose Snap-8 if you want Argireline's longer relative and are comfortable that its efficacy figures come from the supplier.
- Choose Syn-Ake if you want a different target step and a lower use level of 1 to 4%.
- Add Leuphasyl to an Argireline formula if you want the combination with the clearest supporting study.

Frequently asked questions
Is Argireline the same as Acetyl Hexapeptide-8?
Yes. Argireline is Lubrizol's trade name, and Acetyl Hexapeptide-8 is the INCI name you will find on the ingredient list. Older labels and papers may show it formerly listed as Acetyl Hexapeptide-3. When you compare products, check the ingredient list for the INCI name and look for a stated percentage.
What percentage of Argireline should I look for?
Lubrizol lists 2 to 10% for Argireline peptide solution C and 2 to 5% for Argireline Amplified. Those figures refer to the solution as supplied, not pure peptide. The original 2002 study used a 10% cream, so a product disclosing a figure inside Lubrizol's range sits within what has been studied.
Is Snap-8 stronger than Argireline?
The distributor sheet says Snap-8 is about 30% more active than Argireline in lab and human work, but it gives no panel size, duration or concentration. I could not find an independent head-to-head study. Think of Snap-8 as a close relative with supplier-reported advantages, not a confirmed upgrade.
Can I combine Argireline, Snap-8, Syn-Ake and Leuphasyl?
Yes, and many formulas do. They are all water-soluble, and their makers describe three different target steps across the four. The most complete combination data is for Leuphasyl with Argireline, where 43 women saw a larger reduction in line depth with both than with either alone. Keep each one inside its supplier use range.
How long do expression line peptides take to show results?
The human studies reviewed here ran from 4 weeks to 2 months with twice-daily use. Lubrizol claims a change in the look of lines within one week at 2% Argireline, without listing study details. Take photos in the same light at the start and after four weeks to judge your own results.
